When we looked at the gels on Friday morning, we saw that the bands
had developed overnight. This is unusual because the problem with Silver
staining is usually the opposite - silver precipitates too quickly and
the background goes black.
The take home here, is that differenet proteins are represented at different levels in different fractions on the gradient. So the cholesterol rich region has a different profile than the crude homogenate or the pellet. Most of the protein is in the latter and little is found in the CLD fraction. |

The above blot, from a previous year,shows bands on the left in the CLD fractions in the presence of Vanadate. There is a similar gel on the right, made using incubation without vanadate. No bands at all are visible. |
