Protogel

To  make  the  gel  you  need        Running  buffer :    1.5 M  tris  pH 8.8

 

                                                      Stacking  buffer :     0.5 M  tris  pH 6.8

 

                                                     10% w/v  SDS

 

                                                     10% w/v  Ammon. Persulphate ( make fresh on the day )

 

 

                                                      Electrode  buffer      6g Tris ,  28.8 g  Glycine ,  20 ml of  10% SDS

 

 

                                                                                        and  make  up  to 2 L  with  water.

 

 

Protein  loading  buffer :  2x  is  4% SDS , 20% glycerol , 200 mM DTT , 120 mM Tris pH 6.8

 

0.002% Bromophenol blue.

 

 

PVDF  membrane  is  from  Millipore.   One  roll  and  cut  to  size – Immobilon P roll

Transfer  buffer : 5.8 g Tris , 2.9 g Glycine , 3.7 ml 110% SDS , 200 ml Methanol  make up to 1L with water.

 

 

 

 

Blocking  solution :  1%  BSA  in  tris  buffered  saline ( TBS )  containing  0.1%  Tween 20

 

 

 

TBS :  Dissolve  8.0 g  NaCL , 0.2 g KCL , 3.0 g  Tris in  Distilled  water, adjust  to  pH8  with  HCL  and  make  up  to  1L.  Can  be  autoclaved  but  may  not  be  needed  for  this  practical  unless  you  want  to make  lots  and  store  it  for  the 3  weeks.  Store  at  R.T.

 

 

ECL  protocol     stocks        Luminol  from  ICN  195038  or  Fluka  09253 ,  make  a  250mM  stock  soln.  Dissolved  in  DMSO.

 

Indophenol   from  Sigma  1 – 5763  make a  400mM stock dissolved  in  DMSO

 

Glycine – NaoH  buffer  :  0.5M glycine  in  water , pH  to  9.6  with  NaoH

 

Hydrogen  peroxide  30%w/v  (  9.8M )  bought  from  Sigma  or  BDH , goes  off  so  must  be  bought  in  fresh.

 

Developer            2.0 ml    Luminol  stock

 

                             10.0 ml  Indophenol  stock

 

                            100 ml   Glycine – NaoH  buffer    make  to  1000ml  with  water

 

                            Store  in  dark  at  4 C.  Just  before  use  activate  the  reagent  by  adding  36mL  of  Hydrogen  Peroxide  to  20 ml  stock  developer.

 

 

Chemiluminescence  film  :  Kodak  Biomax  light  from  Sigma. (  Z37349 – 4 )

 

                                               We  get  the  35 x  43 cm  box  and  cut  them  if  we  need  smaller  bits.

 

                                               Get  the  biggest  film  size  that  will  fit  the  cassette , it  should  last  for  years  so  buy  a big  box.

 

 

Cholesterol  assay  reagent  :   Sigma  401 – 25P         500ml   liquid    10 x 25 ml Pack

 

 

Cholesterol  calibrator  :  Non certified is O.K.  C0284  5ml

 

 

Bradford  concentrate  for  protein  assays  :  Biorad  450 ml    500 – 0006  ready  to  use

 

                                                                         To  make from  scratch  Dissolve 100mg  coomassie brilliant blue G – 250 in 50ml 95% ethanol.  Add 100ml  conc. Phosporic acid and adjust vol. To 200ml with distilled water. Store at 4 C .

 

 

Antibody  :  The  antibody  is  RC20H  from  Transduction  labs.  Order  through  Beckton Dickinson.     Require  108 mL  cat no.  E120H  and  specify  50mg.  Will  keep  at  - 20.
