Notes on SDS PAGE gels

 

 

 

Use 1% Agarose to seal base and edges of gel plates.

 

Add Separating Gel,   c. 6ml per plate,  leaving 0.5 –1cm between top of

 

separating gel,  and where base of comb will be when fitted.

 

Top up with dist. or butylated water to just below top of lower plate.

 

Leave till gel is set.

 

When set,  pour off water or butylated water.  Use tissue to mop up residue.

 

Top up with Stacking Gel,  till level with top of lower plate.

 

Place comb in position.

 

Allow to set.

 

May be stored overnight wrapped in clingfilm in a coldroom.

 

Any cloudy appearance in gel should disappear on warming to room temp.
