• Remind them that although caveolae contain only 2% total cell protein, contain 98% PrP protein ( and src, kinases, siganlling complexes).
• Invite them to sort themselves into 8 groups of roughly 3

Announcements

  Remind them to use wide-bore tip to remove fractions

  To save time and materials we only take every third fraction.

  Explain Vanadate

  Remind them to label bags so that can find them - separate Vanadate plus/minus

  Remind to keep a crude and pellet sample

  Use gloves in preparing kinase assays

  Remember to bag your fractions. You will need them for protein/cholesterol assays

  Do not forget to bag the crude homogenate. You will need very little for tomorrow, so don't think it is empty and throw it away!

Ask students to leave a note of their names on the desk that they use ( this will enable us  to save time on Day 2, since we can have their samples ready for them).

Safety

Fix up those without a lab' coat.  If so, warn that you are doing them a favour and could send them out of lab'.
