Overview
Caveolae are membrane pits, rich in cholesterol, that may behave as distinct "rafts" in the plasma membrane
They contain many signalling complexes.
Isolate caveolae from other membranes on a sucrose density gradient by virtue of their greater buoyant density.
Show that the majority of cholesterol is found at a discrete part of the sucrose gradient.
Set up an in vitro kinase assay in presence of a phosphatase inhibitor and run a western blot with anti-phosphotyrosine antibody on the gradient fractions.
